Title: Two Possessed by Demons

“Set any 2 humans aside until Son of God is played.”

If both players have already played SoG before this TPbD is played, does ability no longer have any affect if it was played post-SoG?

No, the Humans are indefinitely set aside unless some other special ability brings them back.
