I draw Shame during draw phase, and set aside one of my opponent's heroes, and one of my own. They both have two counters on them. During my next phase (prep), can I remove one of the counters?

If you draw the Shame LS during your draw phase, your next phase is upkeep, during which you remove one counter. One your following turn your Hero comes back.
