If during a game an enhancement was used that gave attack and defense points to a character and it stated that the character keeps those points even after the enhancement is discarded, would this effect be negated later in the game if Job's wife was played against that character?

Nope. Gain is an instant ability, which means it is permanent and cannot be negated after the phase they were played in.

What you are talking about there as 'gain' is Increase, of which there are ongoing and instants (see the REG for more).  Instant increase is permanent, and like all instant abilities cannot be interrupted in a phase after it completes, as Kram stated.
