These are both probably horribly unbalanced, but I thought they were fun ideas and I never make cards. Holy Spirit is relatively straightforward: Searching for Son of God has been one of those things that's been suspicious, but I think that with the dominant cap, this is fairly balanced. The restriction is put in place to give the opponent a chance to play something (primarily VP or Mayhem, though something like Sorrow of Mary would work) to eliminate the benefit of the search.
Number of the Beast is built simply as a one-time block against superior numbers. If anything it's underpowered, but getting high numbers can still be beneficial.

I like the flavor of Number of the Beast. Maybe you could make it operate like answered prayer and let the player choose how to distribute 666 points on the enhancement to make it slightly more powerful.

I think that having two Dominant slots for only one Dominant use is not beneficial. Perhaps if it said SoG or NJ it would be a more likely candidate for the slot (since it could have two uses).
Be aware that for your second card, some Christian parents will not want their child to have a card with that number on it. ;)

If you play Holy Spirit I'm gonna hope I haven't already played VP or I'm not playing hand control. But I do like the idea of an auto block with Number of the Beast

I do like this idea, as long as their isn't a permanent "gain" in the abilities like Answered Prayer. Giving Judas Iscariot 672 defense is scary.
I think giving it an optional "Toss", or optionally making all enhancements "Tossed", could be cool as well.
