Could a deck with no defense work in two player? Or maybe with just Uzzah, some LS generation and drawing?

Yes, these are formally known as speed decks, but a "pure speed" deck with minimal defense isn't as viable as it once was. Basically you build a very strong, fast offense and supplement it with a couple evil characters who are either standalones or characters who help draw cards or generate souls.

I have had some success with defensless decks with no evil characters at all.

I think that at this stage, pure defenseless is actually detrimental to a speedy offense. To use RDT's most popular example, almost all of those cards can be applied to help the offense. A Slave generates a soul, Sabbath Breaker draws, Gomer can band to an enemy EC to potentially get it discarded, and Plot can be used to discard defensive characters. Only KoT and Uzzah can't be used to directly benefit the offense.

And there's no better bang you can get for your buck at 50-52 cards. KoT can get a block, and perhaps multiple blocks, for 1 card (especially useful against Genesis early-game). Uzzah can get a guaranteed block against all but Abigail, Moses and Thad for one card. I guarantee you there's no way to make a deck better at 50-52 cards by taking either of them out and putting something else in.

First of all, by defenseless do you mean no "evil" cards at all? My "defenseless" 50 card deck has DON, FA, Burial and Martyr, and then Am Slave, Malchus, the Gibeonite guy that becomes a lost soul, and Assyrian Survivor. It also has Hopper, Woman at the Well, and Harvest Time for even more Lost Soul generation. I also removed all the "speed" cards that make me draw cards, because this is what I DON'T want to do, and I made it a search deck instead (Stone Pillar at Bethel, Birth Foretold, Wheel within a Wheel, Priests of Christ, etc). It has the ability to generate a ton of lost souls, and the theory is that I'll get to 5 before my opponent.
It's incredibly risky, and a couple cards KILL it, but it is fun to try. Sadly it wins more game than it loses. (I say "sadly" because I think certain offenses are still overpowered.)
Basically, I think "defenseless" is less viable now than it used to be, but it can still win.

Does his ability to self-capture work if you don't have those cards in your discard pile? If it does then he is definitely going back in. Thanks again.

He can self-capture, even without a Genesis Character in the discard pile. This is a case of "do as much as you can"
